Can one prepare suppos based on Trileptal tablets?

What is the stability of Suppo & #039; s. Can we use Novata B as a filler?

Answer

The first question we have to ask is: "Is there rectal absorption of the active ingredient?"

the literature mentions two articles, which strongly question this.

  1. Relative bioavailability, metabolism and tolerability of rectally administered oxcarbazepine suspension.
    CONCLUSIONS: Monohydroxy derivative bioavailability following rectal administration of oxcarbazepine suspension is significantly lower than following oral administration, most likely because of poor oxcarbazepine water solubility. It is unlikely that adequate monohydroxy derivative concentrations can be achieved with rectal administration of diluted oxcarbazepine suspension.

  2. Rectal Absorption of oxcarbazepine, van der Kuy PH, Koppejan EH, Wirtz JJ. Laurentius Hospital, Roermond, The Netherlands.
    Physicians regularly challenge the hospital pharmacy departments to find alternative routes for the administration of drugs, which can & #039; t be withhold, e.g. anti-epileptic drugs. In Our hospital we were confronted with the question whether it was possible to administer Oxcarbazepine rectally. In the present report data on the absorption of rectally administered oxcarbazepine is presented. No therapeutic blood levels were attained after rectal administration. Administration via the oral route, however, gave within the same period of time a therapeutic blood level. It is concluded that the absorption after rectal administration of oxcarbazepine at least in this dose and frequency used is too low to justify application in practice.
