Coca ï ne collyr 10% eye drops?

For the diagnosis of Horner & #039; s syndrome, our ophthalmologist wishes to use a 10% cocaine collyr. We prepared our 4%-IgE Collyr previously, buffered with boric acid, and conserved with Hydroxybenzoates, but the latter do not seem to be optimal for the tolerance. Do any of you have an active and stable formulation for these heavily hypertonic eyedrops?

Answer

The efficacy of this eyedrop will increase with pH increase while and stability and solubility decreases. The pH does not exceed the value six best. Due to the much higher concentration on cocaine I suspect that the amount of borax, present in the 4% composition, will need to be increased. Perhaps the boric acid can completely omitted due This is a bit of testing.
as a preservative, you only have a choice between benzalkonium chloride and the non-ionogenic products. Benzalkonium chloride is strongly discouraged in combination with local anesthetics. The NIPA esters are not a so bad choice considering the occurrence of allergic reactions is not frequent anyway.
