Please advice regarding following sterile eye ointment atropine sulfate 300mg Unguentum oculare Ph. Belg. V AD 30g

  1. question: How to get to this regulation? Personally, I find the prescribed quantity too large. Tenother the available tubes, which are suitable for eye ointments, can only contain 10g ointment.
  2. for the general purpose of the preparation of an eye ointment I would like to refer to the answer to the question about advice regarding the preparation of a sterile eye ointment.  .
  3. since I suspect that atropine sulfate is not sterilizable by dry heat as such, and given the small amount, I would suggest dissolving the atropine sulfate in an aqueous solution of boric acid. These we would also use for eye drops with atropine sulfate.
  4. Prepare a small excess (12g) ointment, in order to be able to deliver 10g. We can easily process 1.2 ml of water in 12g eye ointment. This means that we need to prepare a 10% solution. To compensate for loss in filtration, we prepare 3ml 10% atropine sulfate solution in a 1.5% boric acid solution.
  5. weigh the quantities needed for 10.8 g eye Ointment Base and apply it in a small beaker, which is covered with the help of aluminum foil. Determine the total weight: Eye Ointment base + covered receptacle sterilize by dry heat, allow to cool and place the beaker in the LAF cabinet.
  6. in the LAF cabinet, filter the 3 ml of 10% atropine sulphate solution by sterilizing the sterile receptacle and then measure 1.2 ml with an accurate and sterile syringe.
  7. slowly inject the solution into the sterilized ointment base and mix with a sterile spatula.
  8. transfer 10g ointment into a sterilized eye ointment tube. Close the tube beforehand with the sterilized cap (for sterilization see above question). After filling the tube, it is pinched with a clean sterilized spatula.

The 10g eye ointment, which is filled in the ointment tube, is determined by reweighting. Therefore the determination of the total weight of the eye ointment base + covered receptacle.

Not simple but this should succeed. Create a work plan and a list of all the necessary material and sterilization method.
