Suppositories with a high content of essential oils. Which suppository basis?

We make in pharmacy suppositories with following essential oils -20mg Origanum heracleotitic (Greek Oregano) -20mg Pinus sylvestris (Scots pine) -20mg Origanum Majorana (marjoram -20mg Eucalyptus Radiata -13, 30mg Lavandula Angustifolia (Common lavender) -6, 6mg Thymus Vulgaris We now make it with the Suppocire AS2X, but the STOL point remains too low (we have to pour either liquid and let it solidify in the fridge, or cool it in a cold water bath which ensures too fast solidification). How do I determine exactly the melting point reduction and which suppository basis can I use?

Answer

For volatile oils, which dissolve in the suppository base, we charge a melting point reduction of 0.3 °C for 1% oil; except for chloral hydrate (0.1 °C).

for a number of problem preparations, for which there was a good solution, we are going backwards due to the lack of the appropriate resources. This is valid for the suppositories you cited. A first possibility exists in the use of yellow or white wax to increase the lowered melting point back. Camphor, Guaiacol, eucalyptol and other volatile oils in a concentration of 1% give a smeltpoint reduction of 0.3 °C while for chloral hydrate the value = to 0.1 °C. The finished suppository is best a melting point of +/-35 °C. If the melting point reduction BV is 3 °C, we need to use a mass with melting points 38 °C. The percentage of yellow wax can be read from a chart.

So you calculate the melting point reduction for your suppositories. The finished suppository has quite a melting point of +/-35 ° C. So we need a suppository mass with a stol point of 35 + melting point lowering degrees. To obtain this we add a percentage was added. We'll read this percentage on a chart you can find on
